چکیده

MONG the pyrimidine-requiring mutants of Neurospora is one which exhibits A the requirement only a t temperatures above 31”, in contrast to the more frequently isolated mutants which show an absolute requirement regardless of the temperature of incubation (MITCHELL and HOULAHAN 1947). One of the latter mutants was found to be allelic or quite closely linked to the temperature-sensitive one and to be suppressed by a suppressor of this mutant (HOULAHAN and MITCHELL 1947). More recently a third mutant has been found to be closely linked or allelic to these but is apparently unaffected by the suppressor. This mutant has been shown to give pseudo-wild progeny when crossed to either of the other two (MITCHELL, PITTENGER and MITCHELL 1952). The study which is described here was undertaken in an attempt to learn whether the three mutants are non-allelic.

Genetic studies of ad-8 mutants in Neurospora crassa. II. Interallelic complementation at the ad-8 locus.

N earlier studies with Neurospora, it was believed that with biochemical I mutants the heterocaryon test was a satisfactory test for allelism, based on the expectation that only heterocaryons between nonallelic mutants should be capable of growth on nonsupplemented media (BEADLE and COONRADT 1944). However, experiments by MITCHELL and MITCHELL (1956) suggested that a positive heterocaryon test ...

Recombination in Saccharomyces cerevisiae: a DNA repair mutation associated with elevated mitotic gene conversion.

A mutant haploid strain of Saccharomyces cerevisiae has been isolated that is sensitive to the alkylating agent methyl methanesulfonate at a concentration of 0.01% (vol/vol). The strain also shows sensitivities to x-rays and ultra-violet light, which cosegregate with sensitivity to methyl methanesulfonate as a single gene defect. An analysis of the sensitivity to ultraviolet light indicates tha...

Complementation pattern changes at the pyr-3 locus of Neurospora crassa.

HE pyrimidine-3 (pyr-3) mutants of Neurospora crassa comprise one genetic T locus (SUYAMA, MUNKRES and WOODWARD 1959) , fall into two complementation groups when the very slow growing types are scored as nongrowers (WOODWARD 1962; DAVIS and WOODWARD 1962), are characterized enzymatically by the absence of in vitro aspartate transcarbamylase (ATCase) activity or the presence of ATCase activity i...

Consequences of a suppressor gene effective with pyrimidine and proline mutants of Neurospora.

HIS study was undertaken to find a common factor responsible for the various effects of a suppressor mutation, s, which is able to eliminate the nutritional requirement of certain mutants at the pyrimidine-3 locus and of those at the proline-2 and -3 loci of Neurospora (HOULAHAN and MITCHELL 1947; MITCHELL and MITCHELL 1952). The s gene was not detectable, however, aside from its interaction wi...

Genetic studies of ad-8 mutants in Neurospora crassa. I. Genetic fine structure of the ad-8 locus.

recent years, evidence has accumulated which indicates that genetic re'2mbination occurs not only between different loci but also between alleles within one locus (PONTECORVO 1958). A precise genetic analysis of allelic relationships within one locus is certainly an essential approach to understanding genes, since studies of allelism can help to elucidate both the structure and function of a pa...
